基本介紹: CM-DiI(活細(xì)胞的多代示蹤、固定細(xì)胞長期示蹤) CellTracker CM-DiI為DiI的衍生物,在DiI中加入了具有中度巰基反應(yīng)特性的氯甲基取代物,后者能與細(xì)胞內(nèi)含巰基的肽和蛋白結(jié)合,使得CM-DiI能夠抵抗醛類的固定作用。CM-DiI在細(xì)胞固定、破膜及石蠟包埋的整個(gè)過程中均能很好的保留在細(xì)胞內(nèi),因此,與傳統(tǒng)細(xì)胞膜探針DiI相比較,更適于固定后標(biāo)本的長期示蹤。其紅色/綠色的光譜特征,使其可與綠色探針或者綠色熒光蛋白如GFP一起用于雙標(biāo)實(shí)驗(yàn)。

染色示例 Fig 1.Brain of a zebrafish embryo. CellTracker CM-DiI. A 30 μm–thick section of a zebrafish embryo stained with CellTracker CM-DiI (C7000, C7001) prior to immunohistochemical analysis. A one-day-old zebrafish embryo was immobilized and impaled in the hindbrain with a microelectrode filled with 1 mg/mL CellTracker CM-DiI in 95% ethanol. One day later, the brain was fixed, embedded, frozen in liquid nitrogen and sectioned on a cryostat. After blocking with 0.1% Triton X-100 in PBS containing 2% BSA and 2% normal goat serum, the 30 μm–thick section was incubated with primary anti-glial antibody in conjunction with fluorescein goat anti–mouse IgG antibody. This section was then viewed sequentially through optical filter sets appropriate for rhodamine and fluorescein, and the resulting images were superimposed.
